Current Research Interests

인체의 신경계(nervous system)와 더불어 한 축을 담당하는 면역계(immune system)는 인체의 모든 외부 환경과 직접적으로 맞닿아 있으며, 신경계와 면역계가 서로 유기적인 상호작용을 하여 인체가 환경에 적응하는데에 있어서 필수적인 역할을 하고 있습니다. 심지어 박테리아와 같은 단순한 단세포 생물 조차도 바이러스 감염에 대항할 수 있는 면역체계(효소 시스템)를 갖추고 있습니다. 면역기능으로 인해 비로소 생명체는 환경에 적응할 수 있고 나아가 진화도 할 수 있는 것입니다.  

 

왜 신경-면역 네트워크 연구인가? 

 

면역계와 신경계는 인체에서 가장 중요하고 복잡한 두 시스템으로, 감염이나 외부 변화에 대해 적절히 저항하고 적응하여 항상성을 유지하며 생존을 영위해 가는데 가장 중요한 역할을 담당하고 있습니다. 이 두 시스템을 연구 대상으로 하는 신경과학과 면역학은 그 동안 독자적인 영역으로 연구가 진행되어, 각각의 기능과 질환들에 대한 많은 사실이 규명되는 등, 단일 학문으로서 비약적인 발전을 이루었습니다. 

 

하지만 최근의 연구에 따르면, 그간 독립적으로 간주되어오던 신경계와 면역계 간에 기능적, 구조적으로 상당한 유사성이 있음이 밝혀지고 있는데, 일례로 면역세포들 간에도 신경 시냅스와 유사한 세포 간 시냅스를 형성하여 정보를 주고받는다는 사실이 보고되었으며, 신경계에서만 연구되었던 신경전달물질 (neurotransmitter)이 면역세포 및 면역반응을 조절하고, 면역조절인자들 또한 신경전달물질로 사용된다는 사실이 밝혀졌습니다. 일례로, 뇌수막염, HIV dementia와 같은 감염성 신경질환 뿐만 아니라 알츠하이머, 파킨슨 병, ALS와 같은 여타의 퇴행성 뇌질환에서도 면역염증 반응이 중요한 기능을 하며, 반대로 여러 면역질환들의 진행과 회복이 인체의 자율신경계 및 신경전달물질에 의해서 조절이 된다는 연구결과 또한 보고되었습니다. 

 

이러한 연구 결과들은 기존의 신경질환 및 면역질환 연구가 신경과학이나 면역학만의 독립적인 연구만으로는 그 기전 규명에 한계가 있으며, 따라서 그 질환의 본태를 온전히 이해하기 위해서는 신경계와 면역계의 유기적인 상호작용, 즉 네트워크라는 총체적인 관점 (holistic view)에서 접근하고 연구해야만 한다는 사실을 보여주고 있습니다. 

 

우리 실험실의 주요 관심사는 크게 세 가지 입니다. 

 

첫 번째는 바로 통증(pain) 입니다. 생리적 통증 (physiological pain)은 유해한 외부자극 또는 생존에 위험이 되는 환경으로부터 인체를 보호하는 일종의 경보시스템으로서 생존을 위해 불가결한 기능 중 하나입니다. 병리학적 통증 (pathological pain)은 조직손상, 감염, 염증반응 등의 이유로 인해 발생되며, 일반적으로 손상된 조직의 치유와 함께 자연적으로 소멸됩니다. 하지만 만성화된 병리학적 통증은 그 발병원인이 제거되었음에도 불구하고 계속적으로 발생되어 환자의 삶의 질을 크게 떨어뜨리며 또한 정상적인 경제 및 사회활동을 저해합니다. 따라서 만성통증의 발병기전을 이해하는 것은 통증조절을 위한 약물개발을 위해 필수적이며, 사회적 그리고 경제적으로도 중요한 일입니다. 

 

두 번째 연구주제는 가려움증(itch)입니다. 아토피와 같은 심각한 가려움증은 삶의 질을 현저하게 떨어뜨립니다. 기존의 가려움증에 대한 연구는 신경세포와 그 신호전달에 집중되어 왔지만 최근에는 중추신경계(CNS;central nervous system)에 있는 교세포가 핵심 역할을 할 것이라는 실마리가 하나 둘 씩 제시되고 있으며, 실제로 중추신경계에는 신경세포보다 교세포가 많이 존재하고 있습니다. 이들은 신경세포의 신호전달 및 시냅스 유지 및 제거에 중추적인 역할을 하고 있습니다. 우리 실험실에서는 기존에 신경세포에 맞춰져 진행되어왔던 가려움증에 대한 이해를 신경교세포 관점으로 확대하여 연구하고 있습니다.  

 

통증이나 가려움증이 오랫동안 지속될 경우에 신체적인 고통과 더불어 과연 정신적인 질환을 유발할까?  

 

우리는 우울증(depression) 연구를 통해서 이 가설을 검증하고자 합니다. 만성 통증환자 또는 가려움환자들이 우울증을 겪는다는 여러 임상연구결과들이 보고되었고, 여러 가지 조건에 대해 우울증에 관여하는 많은 유전자들도 밝혀졌습니다. 통증 및 가려움 생쥐모델과 더불어 유전자적중생쥐를 이용하여 생쥐의 우울행동패턴을 분석하고, 이러한 행동패턴을 유발하는 뇌의 신경세포와 교세포의 역할을 밝혀내어 통증 및 가려움증과의 연관성에 대한 다리를 하나씩 놓아보고 있습니다. 

 

우리 실험실의 세 가지 연구 주제는 독립적이지 않습니다. 서로 긴밀하게 연관되어 있으며, 하나의 목적을 가지고 있습니다. 다양한 면역질환과 더불어 이로 인해 발생하는 정신질환에 미치는 영향을 척추(spinal cord)에서 뇌(brain)까지 이르는 종합적인 분석 및 연구를 통해 신경면역계의 새로운 패러다임을 제시하고 그 발전에 기여하고자 합니다.
